Microdosing psilocybin has gone from fringe Silicon Valley biohack to mainstream wellness trend. But between the hype and the skepticism, what do we actually know? This guide breaks down the two most popular protocols, what the research says, and how to do it as safely as possible if you choose to try.

What Is Microdosing?

Microdosing means taking a sub-perceptual dose of a psychedelic. You shouldn't feel "high" or experience visuals. The goal is subtle shifts in mood, focus, or creativity over time. A typical psilocybin microdose ranges from 0.05 to 0.3g of dried mushrooms.

If you're noticing significant perceptual changes, your dose is too high. Scale back.

The Fadiman Protocol

Developed by Dr. James Fadiman, this is the most widely used microdosing schedule. It's simple and conservative.

Schedule:

  • Day 1: Microdose day
  • Day 2: Transition day (no dose, observe aftereffects)
  • Day 3: Rest day (no dose, return to baseline)
  • Day 4: Microdose day (cycle repeats)

You follow this pattern for 4 to 8 weeks, then take a 2 to 4 week break to assess.

The logic is straightforward. The off days prevent tolerance buildup and let you compare "on" and "off" states. Many people journal their mood, energy, and productivity to track changes.

The Stamets Stack

Paul Stamets, the mycologist, proposed a more aggressive protocol that combines psilocybin with two other compounds.

The stack:

  • Psilocybin: 0.05 to 0.2g dried mushrooms
  • Lion's mane mushroom: 50 to 200mg
  • Niacin (vitamin B3): 100 to 200mg

Schedule:

  • Days 1 to 4: Stack daily
  • Days 5 to 7: Off

Repeat for 4 weeks, then take 2 to 4 weeks off.

Stamets theorizes that niacin (which causes a harmless flushing sensation) helps distribute psilocybin and lion's mane to peripheral nerves, enhancing neurogenesis. Lion's mane has its own body of research suggesting neuroprotective properties. The combination is speculative but intriguing.

Important note: the niacin flush can be uncomfortable (skin reddening, tingling, warmth). Start with a lower niacin dose if you're not used to it.

What the Research Actually Shows

Let's be honest about the state of the science.

What we have evidence for:

  • Several small RCTs (randomized controlled trials) suggest microdosing may improve mood, emotional regulation, and creative thinking.
  • Self-report surveys consistently show perceived benefits in well-being, focus, and reduced anxiety.
  • A 2022 study in Nature Medicine found that psilocybin (at therapeutic doses, not microdoses) showed significant antidepressant effects.

What we don't know:

  • Most microdosing studies are small, short-term, and struggle with blinding (participants often guess correctly whether they received psilocybin or placebo).
  • The placebo effect is enormous. Some studies show that expectation alone accounts for much of the reported benefit.
  • Long-term cardiac effects are a real concern. Psilocybin is a 5-HT2B receptor agonist. Chronic 5-HT2B agonism is associated with cardiac valve fibrosis (this is why fenfluramine was pulled from the market). We don't know if microdosing levels of psilocybin carry this risk, but it hasn't been ruled out.
  • Optimal dose ranges aren't established. What works for one person may be too much or too little for another.

Bottom line: microdosing might help some people, but it's not a miracle, and the science isn't settled. Approach it as an experiment, not a prescription.

How to Prepare Your Microdoses

Consistency matters. Different mushroom caps and stems have wildly different psilocybin concentrations.

Steps:

  1. Dry your mushrooms completely. They should snap, not bend.
  2. Grind them to a fine powder using a coffee grinder. This evens out the potency distribution.
  3. Weigh individual doses on a milligram scale (0.001g precision).
  4. Fill capsules for consistent, easy dosing. Size 00 or 0 capsules work well.
  5. Store in a cool, dark, airtight container. Psilocybin degrades with heat, light, and moisture.

If you don't grind and capsule, you're guessing. Two mushrooms from the same batch can vary significantly in potency.

When NOT to Microdose

Do not microdose psilocybin if you:

  • Are on lithium (combination can trigger seizures and is considered dangerous)
  • Have a personal or family history of psychosis or schizophrenia
  • Are on tramadol (lowers seizure threshold)
  • Are pregnant or breastfeeding
  • Have a heart condition (due to the 5-HT2B concern mentioned above)

If you're on SSRIs, be aware that they may blunt the effects of psilocybin. Don't increase your dose to compensate. Talk to a doctor if possible.

Starting Your Protocol

If you've decided to try microdosing:

  1. Start with the lowest dose (0.05 to 0.1g) and work up slowly. You want sub-perceptual.
  2. Pick one protocol (Fadiman or Stamets) and stick with it for at least 4 weeks before judging.
  3. Keep a journal. Track mood, sleep, focus, creativity, and any side effects. Without data, you're just vibing.
  4. Take breaks. Both protocols include off-days and off-weeks for a reason. Tolerance, 5-HT2B concerns, and integration all benefit from rest periods.
  5. Don't microdose every day. This defeats the purpose and increases risk.
  6. Have realistic expectations. Microdosing is not a shortcut to mental health. It's one tool among many.

FAQ

What's the difference between the Fadiman and Stamets protocols?

Fadiman uses a simple 1-day-on, 2-days-off cycle with psilocybin only. Stamets uses a 4-days-on, 3-days-off cycle and adds lion's mane mushroom and niacin to the mix. Fadiman is more conservative and better studied. Stamets is more experimental.

Can I microdose while on antidepressants?

SSRIs and SNRIs may reduce the effects of psilocybin, making microdosing less effective. Do not stop your medication to microdose. If you're considering combining the two, talk to a doctor. Never microdose on lithium or MAOIs.

How do I know if my dose is too high?

If you notice visual distortions, significant mood swings, difficulty concentrating, or feel noticeably altered, your dose is too high. A proper microdose should be barely noticeable. Scale back by 0.05g and reassess.

Will microdosing show up on a drug test?

Standard workplace drug tests (5-panel or 10-panel) do not screen for psilocybin. However, specialized tests can detect it. The detection window is short (typically under 24 hours in urine), but don't rely on this if testing is a concern.

Is microdosing legal?

In most countries, psilocybin is a controlled substance, making both possession and microdosing illegal. Some jurisdictions (parts of the US, the Netherlands with truffles, Jamaica, Brazil) have decriminalized or allow it in certain contexts. Know your local laws before proceeding.

How long should I microdose before expecting results?

Most protocols recommend 4 to 8 weeks before evaluating. Some people notice subtle shifts within the first week. Others feel nothing. If after 8 weeks you haven't noticed any benefit, microdosing may not be for you, and that's fine.
